**Background**
Parkinson’s disease is a progressive neurodegenerative disorder characterized by the loss of dopaminergic neurons and the accumulation of misfolded $\alpha$-synuclein protein aggregates. The toxicity associated with $\alpha$-synuclein leads to severe impairment of intracellular trafficking, particularly affecting the transport between the endoplasmic reticulum (ER) and the Golgi apparatus. Restoring these trafficking pathways is a critical therapeutic goal to prevent neuronal death and ameliorate the symptoms of early-onset Parkinson’s disease. The small GTPase Rab1a plays a pivotal role in regulating ER-to-Golgi trafficking and mediating endosomal events involving the E3 ubiquitin ligase Rsp5/Nedd4. Consequently, targeting the Rab1a pathway has emerged as a promising strategy for neuroprotection. In this context, we will introduce a neuroprotectant that targets Rab1a – NAB2.
**Definition**
NAB2 is a neuroprotectant that selectively binds to the GDP-bound form of the small GTPase Rab1a, with $\alpha$-synuclein as its primary target.
**Mechanism of Action**
According to the NAB2 description, this compound acts by increasing the expression of Rab1a, which in turn regulates essential ER-to-Golgi trafficking. Furthermore, NAB2 stimulates the ubiquitination of related proteins in a Nedd4-dependent manner. By enhancing these processes, NAB2 is able to rescue $\alpha$-synuclein-associated trafficking defects that are characteristic of early-onset Parkinson’s disease. Detailed NAB2 technical information indicates that the compound’s ability to stabilize Rab1a helps protect multiple cell types from the cytotoxic effects of $\alpha$-synuclein.
**In Vitro Studies**
The NAB2 biological activity has been demonstrated through its ability to protect various cell types from $\alpha$-synuclein toxicity. In vitro studies show that by targeting the GDP-bound form of Rab1a, NAB2 effectively mitigates the cellular stress and trafficking failures induced by $\alpha$-synuclein. This mechanism ensures the maintenance of proteostasis and prevents the neuronal degeneration typically observed in Parkinson’s disease models. In conclusion, NAB2 is a potent neuroprotectant that rescues $\alpha$-synuclein-associated toxicity by modulating the Rab1a/Nedd4 pathway.
