**Background**
Cancer remains one of the most challenging health crises globally, characterized by uncontrolled cell proliferation and the ability to evade programmed cell death. Among the various malignancies, breast cancer, lung cancer, melanoma, and glioma are particularly aggressive and often require the development of novel therapeutic agents to overcome drug resistance and improve patient outcomes. Metal-based complexes have long been utilized in oncology, but there is a continuous need for compounds with higher potency and lower systemic toxicity. N-heterocyclic carbene (NHC) complexes, particularly those involving copper, have emerged as promising candidates due to their stability and diverse biological activities. In this context, we will introduce a potent anticancer agent – Chloro[1,3-bis(2,6-diisopropylphenyl)imidazol-2-ylidene]copper(I).
**Definition**
Chloro[1,3-bis(2,6-diisopropylphenyl)imidazol-2-ylidene]copper(I) is an NHC copper complex that acts as an anticancer agent, exhibiting potent cytotoxic activity against multiple cancer cell lines.
**In Vitro Studies**
According to the Chloro[1,3-bis(2,6-diisopropylphenyl)imidazol-2-ylidene]copper(I) description, this compound demonstrates significant inhibitory effects on the proliferation of various human cancer cells. In Chloro[1,3-bis(2,6-diisopropylphenyl)imidazol-2-ylidene]copper(I) in vitro assays conducted over a 72-hour incubation period, the complex showed high potency across several models. Specifically, it inhibited the proliferation of MCF-7 breast cancer cells with an IC50 value of 0.15 μM and A549 lung carcinoma cells with an IC50 of 0.04 μM. Furthermore, it inhibited the growth of SK-Mel-28 melanoma cells with an IC50 of 0.045 μM and HS-683 glioma cells with an IC50 of 0.65 μM. Notably, in cytotoxicity assays against A549 cells, this complex exhibited a lower IC50 than the reference compound Etoposide, highlighting its superior Chloro[1,3-bis(2,6-diisopropylphenyl)imidazol-2-ylidene]copper(I) biological activity. In conclusion, Chloro[1,3-bis(2,6-diisopropylphenyl)imidazol-2-ylidene]copper(I) is a potent NHC copper complex with broad-spectrum cytotoxic activity against breast, lung, melanoma, and glioma cancers.
